What does a spectrophotometer measure

A spectrophotometer measures the amount of light a substance absorbs or transmits at specific wavelengths, allowing determination of concentration and chemical properties.

Core Measurement Principles

A spectrophotometer is an optical instrument that quantifies how much light passes through (transmittance) or is absorbed by a sample (absorbance) at particular wavelengths of the electromagnetic spectrum. When light interacts with a sample, molecules absorb energy at characteristic wavelengths, which reduces the intensity of light reaching the detector. The remaining light is measured, and the absorbance is calculated, often using the Beer-Lambert Law, which relates absorbance to the concentration of the light-absorbing substance.

Key Measurements

Absorbance (A): The logarithmic measure of light absorbed by the sample. Higher absorbance indicates more light is absorbed, which correlates with higher concentration of the substance. Transmittance (T): The percentage of light that passes through the sample.

For example, if 80% of light passes through, the transmittance is 80%. Reflectance (optional): Some spectrophotometers can measure the fraction of light reflected from a solid or opaque sample.

Wavelength Ranges

Spectrophotometers can operate across various ranges of the electromagnetic spectrum: UV-visible: 185–700 nm, commonly used for chemical and biochemical analysis. Infrared (IR): 700–15,000 nm, used for molecular structure and material studies. Other ranges: Modern instruments can also measure X-ray, microwave, or other wavelengths depending on the design.

Applications

Chemical analysis: Determining concentrations of solutions, including enzymes, proteins, or colored compounds. Clinical diagnostics: Measuring blood or tissue components. Material science: Evaluating transparency, color, or reflectivity of solids and liquids.

Research: Studying reaction kinetics, equilibrium constants, and molecular interactions. By measuring how light interacts with a sample, spectrophotometers provide both qualitative information (identity of a substance) and quantitative information (concentration or amount of substance) across a wide range of scientific and industrial applications.
